How the math works

What is peptide reconstitution?

Reconstitution is dissolving a lyophilized (freeze-dried) peptide in a sterile diluent to produce a solution of known concentration. The mass of peptide in the vial divided by the volume of diluent added gives the working concentration in mg/mL.

How do I calculate concentration?

Concentration (mg/mL) = peptide mass (mg) ÷ diluent volume (mL). A 10 mg vial reconstituted with 2 mL of diluent yields 5 mg/mL. Adding more diluent lowers the concentration; it does not change the total mass in the vial.

How do I work out an aliquot volume?

Volume (mL) = target mass (mg) ÷ concentration (mg/mL). At 5 mg/mL, withdrawing 0.5 mg of material requires 0.1 mL of solution.

Does the diluent volume change how much peptide is in the vial?

No. The vial contains a fixed mass regardless of how it is reconstituted. Diluent volume only sets the concentration, and therefore the volume needed to transfer a given mass.

How should reconstituted material be stored?

Reconstituted peptide solutions are generally held refrigerated and protected from light, and are less stable than lyophilized powder. Consult the batch Certificate of Analysis and your own laboratory's handling protocols.
